Mine would have to be Gears of war.
The only reason I haven't sold my 360 is Gears of War.
Definitely Mass Effect. I thought Gears of War 3 was way too much filler and seemed like a waste of time considering it was supposed to be this hard-hitting action-packed conclusion to one of the most epic series of all time, but I didn't feel that. Also no substantial changes in multiplayer overall.
Mass Effect. Though, they did port number 2 to the PS3. It is still the defining series for the Xbox 360. Halo was good but then it got incredibly absurd in the second and third one. I sure hope Halo 4 can make up for all the lost ground.
As far as Gears goes, they were all awesome. The story mode left a lot to be desired though. Yes, they tried to make you care about the whole squad but there was a lot of "bro talk" (a la Army of Two). It wasn't necessarily bad but still could of been sooo much better.

Probably Assassin's Creed. But Halo and Gears are a very close 2nd and 3rd.
I have to say Mass Effect, but some very close others are Halo, Forza, and Fable.
Assassins Creed all the way.
Fable overall (though I am willing to admit fable 3 was a step backwards). I also really enjoy the Forza series.
The best series I've played would have to be Mass Effect. I actually haven't done much of Gears of War, but should give it a shot.

halo series and borderlands (i know borderlands is on both platforms but i just like it better on xbox360) only reason i have a ps3 is for god of war series anyways
Mine is probably Gears of war as well but Halo is making a strong push to over take it.
The Mass Effect series was, by far, the most satisfying and emotionally/mentally charged gaming experience of my life. It remains one of the most amazing science fiction stories in which I will ever take part.
The Elder Scrolls, although Mass Effect is quickly becoming a favorite.
